The protocol at a glance
Trials dose kisspeptin-54 by body weight (nmol per kg), not as a fixed amount. Repeated twice-daily dosing made the effect fade in women with missed periods.
| Study | Amount of kisspeptin-54 |
|---|---|
| Human dose for general use | Not established |
| IVF egg-maturation trigger, one shot under the skin (60 women, 2015) | 3.2 to 12.8 nmol per kg; later trials used 9.6 nmol per kg |
| Women with missed periods, under the skin twice a day for 2 weeks | 6.4 nmol per kg |
| Nasal spray, healthy adults and women with missed periods (2025) | 12.8 nmol per kg |
| Test dose into a vein, men with and without a hormone-gene disorder (2021) | 6.4 nmol per kg |

Kisspeptin-10 (a shorter form) was given into a vein as 0.3 mcg per kg in one dose, or 4 mcg per kg per hour as an 11-hour drip, in men with type 2 diabetes (PMID 23153270).
Community reports commonly describe 100-200 mcg every other day.
What people say they do, collected from community discussions. Not tested in a trial; shown so you can compare it with the evidence above.
Mixing
No mixing table here: this one is used as a capsule, nasal spray or skin product, or has no human amount to calculate from. The general mixing calculator works for any vial.
What the trials found
Two human studies of kisspeptin-54 under the skin. IVF trigger, 60 women at high risk of overstimulation (Abbara 2015), and 10 women with missed periods given twice-daily shots (Jayasena 2009):
Abbara A et al. J Clin Endocrinol Metab 2015 (PMID 26192876); Jayasena CN et al. J Clin Endocrinol Metab 2009 (PMID 19820030). The women with missed periods still responded to GnRH, so the pituitary gland still worked.
Side effects
- The 2025 nasal-spray study reported no side effects or adverse events.
- With repeated twice-daily shots for 2 weeks, the hormone response faded (desensitization).
- In healthy women, 1 week of twice-daily shots did not stop cycles, but cycles got shorter on average (26.8 vs 28.6 days).
- Long-term safety is not established.
How it works
Brain GnRH cells
Kisspeptin acts in the hypothalamus to release GnRH, which tells the pituitary gland to release LH and FSH.
Effect fades with repeats
Daily or twice-daily dosing over days to weeks reduced the response in women with missed periods.
